Wishing to create a mutant army with formidable combat capabilities, Draxum used Lou Jitsu's DNA paired with his ooze to turn Leo from an ordinary turtle into a mutant. He and his brothers were then stolen from Draxum's laboratory by Lou Jitsu promptly after their mutation and Leo went on to be raised in the sewers of New York City alongside his brothers by Splinter. Eventually, Leo and his brothers met a human girl named April and forged a strong bond with her through the years. In the future of 2044, Leonardo is part of a resistance faction, warring against the aliens called Krang, who had begun an invasion over 20 years prior and started the apocalypse. At some point, Leonardo had taken a young Casey Jones under his wing and mentored him, recruiting him as part of the resistance. Leonardo lost his right arm in a battle against the Krang. Leonardo is the leader and the commander of The Resistance, the last survivors of the apocalypse and Krang invasion. The members of The Resistance swore to save and protect Earth at all costs. The Krang forced the last survivors to hide in caves and eat rats as a last resort.).

However, the situation quickly escalated into chaos, with casualties mounting and blood spilling all around.
